Well, Mixed Myth mentioned a TV character but I have to second her. The Marquis de Carabas! Kinda a nastier version of Zaphod. I likes him.
Captain Jack Sparrow, of course. He really does carry those movies and I consider it a bit of a shame that the movies aren't really good enough to deserve it. I mean, they're fun but Jack's the only really good thing about them.
Han and Indy. Harrison Ford at his subtly comedic best. The T-100 for his cat-like awareness and implacable determination. Mal from Serenity (a bit of a cheat, but legal by the rules laid out herein). Picard from First Contact. Mulan from her self-titled movie.
And, like everyone else, I'm probably forgetting some.

Ha ha Robin, I was so gonna say Wolf too.
Also, Bellboy from "Four rooms". So impressive, it almost made me love that godawful thing.
Mrs Robinson from The Graduate.
Piter Seller's Mr. Qiumby from "Lolita"
Main character from "Sky over Berlin".
Johnny Depp's Ed Wood.
Forest Whitaker's character from "Ghost dog - the way of Samurai"
Jay and Silent Bob. They are quite worn out now, but back when they appeared they were something different.
Jesus from "Jesus Christ Vampire Hunter". Heh heh heh.
